- Identity. Retention time by RP-HPLC against the reference standard, confirmed by ESI-MS. The spectrometer should place the protonated molecule at m/z 774.9 for 1+, against a calculated average mass of 773.9 (C35H55N11O9); the measured mass should agree to within a unit.
- Related substances. Any single related substance not more than 1.0 percent. On this sequence the ones to look for are light products of the tryptophan (kynurenine and hydroxytryptophan).
- Main peak position. On C18 the main peak comes about mid-gradient; the backbone’s Kyte-Doolittle average is 0.14 over 7 residues and the net charge near neutral is about +1.
- Water. Not more than 8.0 percent by Karl Fischer.
- Fill. Labelled peptide content per vial, plus or minus 5 percent by weight.
- Endotoxin. Reported on the certificate by LAL where the line is tested.
